Principle of the MAT Vector® System

The MAT Vector® System is based on a unique concept that Agrobacterium oncogenes can be used as a selectable marker to regenerate transgenic plants and select marker-free transgenic plants.

Oncogenes are generally not used as a selectable marker for plant transformation because the resulting transgenic plants exhibit a serious abnormal phenotype. The MAT Vector®System is designed to remove the oncogenes from transgenic plants after transformation by utilizing a yeast site-specific recombination system, R/RS.  ref. 3,4  Two kinds of MAT Vectors are constructed with the oncogenes. (1) Cytokinin type: the isopentenyl transferase (ipt) gene of A. tumefaciens PO22 is used as a selectable marker. (2) Auxin type: a 7.6 kb EcoR I rol gene fragment from A. rhizogenes NIAES1724. ref. 5,6

Principle of the MAT 
Vector® System

Advantages

In current transformation systems, selective agents (antibiotic, herbicide, etc.) and corresponding resistance genes (selectable marker genes) are used to select transgenic plants. However, these methods have three major problems: (1) the selective agents decrease the ability of plant cells to proliferate and differentiate; (2) the uncertainty of environmental impacts on many selectable marker genes; and (3) difficulties in repeating a transformation to increase the number of the desired genes using the same selectable marker. ref.7  The MAT Vector® System is designed to overcome these difficulties. ref.8,9  The MAT Vector® System provides an alternate approach to regenerating various recalcitrant plant species for transformation, through internal manipulation of the cytokinin-to-auxin ratio, and it also enables the production of marker-free transgenic plants without sexual crosses and seed production for a pyramid of multiple genes into a vegetative propagated crop by repeated transformation.
